DNA EXTRACTION
The video shows a small lump of human dna - extracted from a few drops of capillary blood using only homemade ethanol precipitation protocols - stored in an Eppendorf tube under EtOH.
The yield was satisfying.
-The lysis buffer was an alkaline lysis buffer based on sodium laureth sulphate, Na2EDTA, sodium chloride for protein precipitation; the pH was adjusted using HCl and NaOH. DNA free
water was used.
-the protein precipitation buffer was composed only of Subsilisin A dissolved in ddH20
-the cells were incubated (see here) and heated to allow lysis, then cooled and protein precipitation was allowed. Precipitation was
achieved using cold EtOH and the tube was mixed by inversion. The DNA was washed under EtOH and stored either under ethanol or dissolved in buffer.
-Lysis buffer composition:
- SDS 1%
- Na2EDTA 100mM
- NaCl 100mM
- pH adjusted to 8 using HCl/NaOH
